Use and description:
For the transportation and recovery of microorganisms acc. ISO/DIS 6649. This solution protects organism by peptone and physiological saline osmotic support.
Composition per litre:
Sodium Chloride………………..…………….8.5 g
Peptone..…………..…………………………..1.0 g
Final pH of the ready to use medium: 7.0±0.2
Medium preparation:
Add 9.5 grams of dehydrated culture medium to 1 litre of distilled water. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute and autoclave for 15 min at 121°C. To 10 g test samples add 90 ml of sterile MRD and blend it. Make 10x serial dilutions from the macerate and add 1 ml of it to the chosen medium on 45-50°C. Let it solidify and incubate.
Quality specifications:
Dehydrated medium: homogeneous, white fine powder.
Ready to use medium: colourless clear solution.
Storage:
Dehydrated medium should be stored between 10 to 25°C. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or if the colour has changed from the original.
